The secretory expression of the organophosphorus compound degrading enzyme with industrial and military uses in Bacillus subtilis bacteria

Introduction: Organophosphate is the common name for phosphoric acid esters that are used in various fields, especially in military productions. Considering the production and use of chemical weapon in recent wars, researches for identifying, preventing and treating pollution with these compounds is essential. These compounds have a lot of negative effects on the biological system. Studies have shown that the biodegradation of these compounds is the most effective method for eliminating these compounds. Various enzymes including the opdB enzyme have been identified for the degradation of these compounds.
Method: The opdB gene was amplified by PCR from the genome of Lactobacillus brevis ATCC367. The PCR product was cloned in pWB980 vector. Susceptible B. subtilis WB800 strains were transformed by recombinant vector. Secretion expression of protein in superrich medium was investigated using SDS PAGE technique. The enzyme activity was determined by a spectrophotometeric method using chlorpyrifos as the substrate.
Result: When we analyzed protein fractions with SDSPAGE, a 27 kDa protein band was presented. Enzyme activity was estimated at 10/83 unit/mL.
Discussion: The results show that the recombinant strain is capable of producing an efficient and active secretory enzyme. |
